Lymphocyte subset reference intervals in blood donors from northeastern Brazil Anais da ABC (AABC)
TORRES,ALEX J.L.; CISNEIROS,PATRÍCIA; GUEDES,ROSA; GRASSI,MARIA FERNANDA R.; MEYER,ROBERTO; BENDICHO,MARIA T.; LOPES,TAIS G.S.L.; FÉLIX,GABRIELA; NETTO,EDUARDO M.; BRITES,CARLOS; ABE-SANDES,KIYOKO; BRANDÃO,CLAUDIO; ALCANTARA-NEVES,NEUZA; FREIRE,SONGELI M..
The reference intervals for leukocytes and lymphocytes currently used by most clinical laboratories present limitations as they are primarily derived from individuals of North American and European origin. The objective this study was to determine reference values for peripheral blood B lymphocytes, T lymphocyte subsets (CD4+, CD8+, naïve, memory, regulatory, TCRαβ and TCRγδ+) and NK cells from blood donors in Salvador-Bahia, Brazil. Results: The proportion of included male subjects was 73.7% and the median ages of males (34) and females (35) were found to be similar. Absolute counts total lymphocytes subsets to both gender was 1,956 (1,060-4,186) cells and relative values 34%. The T CD4+ and T CD8+ lymphocytes relative values was 51% (20-62) and 24%...

Measurement of annexin V uptake and lactadherin labeling for the quantification of apoptosis in adherent Tca8113 and ACC-2 cells BJMBR
Hu,T.; Shi,J.; Jiao,X.; Zhou,J.; Yin,X..
Phosphatidylserine (PS) exposure occurs during the cell death program and fluorescein-labeled lactadherin permits the detection of PS exposure earlier than annexin V in suspended cell lines. Adherent cell lines were studied for this apoptosis-associated phenomenon to determine if PS probing methods are reliable because specific membrane damage may occur during harvesting. Apoptosis was induced in the human tongue squamous carcinoma cell line (Tca8113) and the adenoid cystic carcinoma cell line (ACC-2) by arsenic trioxide. Cells were harvested with a modified procedure and labeled with lactadherin and/or annexin V. PS exposure was localized by confocal microscopy and apoptosis was quantified by flow cytometry. The detachment procedure without trypsinization...

Métabolisme énergétique du spermatozoïde d'huître creuse (Crassostrea gigas) ArchiMer
Boulais, Myrina.
The present study aims at describing the energy metabolism of Pacific oyster, Crassostrea gigas, spermatozoon. The main objectives are: (1) describing the kinetic of cellular and biochemical characteristics of Pacific oyster spermatozoon during its movement phase, (2) identifying the metabolic pathways to produce energy and their substrates, (3) determining the involvement of energy metabolism in fertilization success. The results showed the long duration (ranging 24 to 72h) of the movement of Pacific oyster spermatozoon and the maintenance of a high intracellular ATP content until the end of the movement phase. The role of oxidative phosphorylation (OXPHOS) in ATP production was highlighted. Alternative ATP producing pathways might contribute to sustain...

Multigenerational exposure of the microalga Tetraselmis suecica to diuron leads to spontaneous long-term strain adaptation. ArchiMer
Stachowski-haberkorn, Sabine; Jerome, Marc; Rouxel, Julien; Khelifi, Celia; Rince, Maeva; Burgeot, Thierry.
To investigate the ability of microalgae to develop stable, long-term resistance to herbicides, the marine microalga Tetraselmis suecica was exposed to the herbicide diuron (5 µg/L) for a 43-generation exposure period followed by a 12-generation depuration phase. During the first 25 generations, diuron-exposed cultures showed doubling times ranging from 1.95 to 2.6 days, which was 2 to 2.5-fold longer than control cultures. Between generations 25 and 38, during diuron exposure, two out of the three exposed cultures exhibited a spontaneous drop in doubling time. These results provided evidence of culture adaptation to diuron. To assess persistence of the diuron adaptation observed on growth performance, one of the adapted cultures (D3) was maintained for 12...

Neoplasia detection in Macoma balthica from the Gulf of Gdansk: comparison of flow cytometry, histology and chromosome analysis ArchiMer
Smolarz, Kasia; Renault, Tristan; Soletchnik, Patrick; Wolowicz, Macciek.
A flow cytometry protocol was applied for the detection of neoplasia in Macoma balthica L. from the Gulf of Gdansk (Baltic Sea, Poland). A simple method, based on an osmotic shock, was used to permeabilise gill cells. The cytometric pattern of normal clams consisted of 2 peaks, a major peak B and a smaller peak C. The cytometric pattern of affected clams consisted of 2 peaks named B' and C'. Two parameters were used to define the stages of abnormalities in M. balthica clams based on the percentage of cells in peaks B, C, B' and C' and on the ratio between the fluorescence value of peaks B, C, B' and C' in all individuals. Three stages of neoplasia were clearly distinguished by flow cytometry considering peak C'. Stage 1 was characterised by a major...

Nuclear DNA content and chromosome number in Brachiaria spp. genotypes Rev. Ciênc. Agron.
Timbó,Ana Luiza de Oliveira; Pereira,Roselaine Cristina; Souza Sobrinho,Fausto; Davide,Lisete Chamma.
Breeding programs for Brachiaria spp. use both intraspecies and interspecies crosses between sexual and apomictic plants in order to obtain new cultivars with the desired characteristics. As there are different ploidy levels both within and between species of this genus, it becomes necessary to evaluate the genotypes used in breeding programs, as a guide to breeders when adopting crossing strategies. In this work, DNA content and chromosome number were determined in order to characterise ploidy levels in Brachiaria spp. genotypes. In the analysis of 15 genotypes, DNA content varied with the ploidy levels (2x, 3x and 4x), and between species and/or taxon. The average DNA content was 1.74 pg (2x) in B. ruziziensis, 3.74 pg (4x) in B. decumbens and 3.52 pg...

Nuclear DNA content and karyotype of Rosewood (Aniba rosaeodora) Genet. Mol. Biol.
Contim,Luis Antônio Serrão; Carvalho,Carlos Roberto de; Martins,Franciele Alline; Freitas,Danival Vieira de.
Rosewood (Aniba rosaeodora Ducke, Lauraceae) is ecologically and economically important to the Amazon region. As a consequence of its economic importance, rosewood populations have been decimated in the Amazon forest. Species of nine genera of the Lauraceae family have characterized karyotypes with n = x = 12 chromosomes in the gametophytic phase but the genus Aniba is one of the least studied Lauraceae genera with a previously undescribed genome. We used cytogenetic techniques to determine that the A. rosaeodora karyotype contained 12 pairs (2n = 24) of relatively small submetacentric chromosomes with lengths ranging from 1.34 to 2.25 mum and a nucleolar organizer region (NOR) in the short arm of chromosome 7. Flow cytometry gave 2C = 2.32 pg of DNA,...

Pacific oyster (Crassostrea gigas) hemocyte are not affected by a mixture of pesticides in short-term in vitro assays ArchiMer
Moreau, Pierrick; Burgeot, Thierry; Renault, Tristan.
Pesticides are frequently detected in estuaries among the pollutants found in estuarine and coastal areas and may have major ecological consequences. They could endanger organism growth, reproduction or survival. In the context of high mortality outbreaks affecting Pacific oysters, Crassostrea gigas, in France since 2008, it appears of importance to determine the putative effects of pesticides on oyster susceptibility to infectious agents. Massive mortality outbreaks reported in this species, mainly in spring and summer, may suggest an important role played by the seasonal use of pesticides and freshwater input in estuarine areas where oyster farms are frequently located. To understand the impact of some pesticides detected in French waters, their effects...

Phytoplankton dynamics in the southern New Caledonian lagoon during a southeast trade winds event ArchiMer
Neveux, Jacques; Lefebvre, Jean-pierre; Le Gendre, Romain; Dupouy, Cecile; Gallois, Francis; Courties, Claude; Gerard, Philippe; Fernandez, Jean-michel; Ouillon, Sylvain.
During the Echolag cruise (13 February-8 March 2007), the effect of 16-30 knot trade winds on abundance, community structure and dynamics of phytoplankton was investigated in the southern part of the New Caledonian coral reefs and surrounding oceanic waters. In this area, the coral reefs form two horn-like structures (hereinafter referred to as 'horn reefs'), oriented south-east, and separated by a relatively deep valley. Three repeated samplings at one week intervals of a 32 station array showed that trade winds induce surface water cooling and significant enrichment of mid- and bottom waters above the reef shelf. This enrichment appeared as the result of a general rise of the oceanic thermocline and nitracline due to the combination of trade winds and...

Pollutant effects on Pacific oyster, Crassostrea gigas (Thunberg), hemocytes: Screening of 23 molecules using flow cytometry ArchiMer
Gagnaire, Beatrice; Thomas Guyon, H; Burgeot, Thierry; Renault, Tristan.
The shellfish industry is an important economic activity in France, occurring mostly in estuarine zones subject to pollution due to anthropogenic activities. The harmful effects of pollutants on species inhabiting these estuarine zones are not well known. Among marine species, bivalve mollusks---particularly Pacific oyster, Crassostrea gigas-may serve a model of interest. The species is sedentary and filter-feeding, which favors bioaccumulation of pollutants in their tissues. Oysters may be suitable for studies on disturbance by pollutants of physiological activities, among which defense mechanisms are poorly documented in bivalves. In this study, effects of pollutants on hemocyte functions were monitored in Pacific oyster, C. gigas. Hemocytes were exposed...

Polystyrene microbeads modulate the energy metabolism of the marine diatom Chaetoceros neogracile ArchiMer
Seoane, Marta; González-fernández, Carmen; Soudant, Philippe; Huvet, Arnaud; Esperanza, Marta; Cid, Ángeles; Paul-pont, Ika.
Due to the growing concern about the presence of microplastics (MP) in the environment, the number of studies evaluating the toxicity of these small persistent particles on different marine species has increased in recent years. Few studies have addressed their impact on marine phytoplankton, a subject of great concern since they are primary producers of the aquatic food web. The aim of this study is to unravel the cytotoxicity of 2.5 μg mL−1 unlabelled amino-modified polystyrene beads of different sizes (0.5 and 2 μm) on the marine diatom Chaetoceros neogracile. In addition to traditional growth and photosynthesis endpoints, several physiological and biochemical parameters were monitored every 24 h in C. neogracile cells by flow cytometry during their...

Possible bloom of free trichomes in the Bay of Marseille, NW Mediterranean Sea: an anomaly evidenced by flow cytometry ArchiMer
Zhao, Li; Denis, Michel; Barani, Aude; Beker, Beatriz; Mante, Claude; Xiao, Tian; Gregori, Gerald.
The composition of ultraphytoplankton (10 m) sampled in surface waters (1 m depth) was investigated during summer 2010 in the Bay of Marseille (NW Mediterranean) using flow cytometry. In addition to groups of Synechococcus, Prochlorococcus, picoeukaryotes and nanoeukaryotes, an unidentified additional cluster was observed. The particles forming this cluster had a high abundance (7 10(4) events mL(1)), with both orange and red fluorescence intensities like Synechococcus, but with a size signal larger than that of 2 m fluorospheres (beads). These unknown particles were sorted out by flow cytometry and then observed by epifluorescence microscopy: they appeared to be chain-forming microorganisms, just like trichomes of some diazotroph cyanobacteria with one...

ABSTRACT Agapanthus (Agapanthaceae) has 10 species described. However, most taxonomists differ respect to this number because the great phenotypic plasticity of the species. The cytogenetic has been an important tool to aid the plant taxon identification, and to date, all taxa of Agapanthus L'Héritier studied cytologically, presented 2n = 30. Although the species possess large chromosomes, the group is karyologically little explored. This work aimed to increase the cytogenetic knowledge of Agapanthus africanus (L.) Hoffmanns by utilization of chromosome banding techniques with DAPI / CMA3 and Fluorescent in situ Hybridization (FISH). In addition, flow cytometry was used for determination of DNA content and the percentage of AT / GC nitrogenous bases....

In the last decades, shellfish farming and in particular ostreiculture have developed in a significant way in France. Marennes-Oleron (Charente-Maritime) basin is the first French producer of oysters. But this area is also subjected to many recurring pollution, brought mainly by the Charente river. The recrudescence using of herbicides in agriculture including atrazine implies the transfer towards the aquatic environment of new pollutants in estuarine areas. Moreover industrial rejections bring many heavy metals in these same estuarine areas. It is appropriate of wondering on harmful effects of all these pollutants on the animals living in these areas, particularly oysters. Bivalve molluscs have been postulated as ideal indicator organisms because of...

Reduction of adhesion properties of Ruditapes philippinarum hemocytes exposed to Vibrio tapetis ArchiMer
Choquet, Gwenaelle; Soudant, P; Lambert, C; Nicolas, Jean-louis; Paillard, C.
Vibrio tapetis is the causative agent of brown ring disease (BRD), which affects a species of clam, Ruditapes philippinarum. After incubation with V. tapetis, hemocytes lose filopods and become rounded, indicating cytotoxic activity of the bacterium. To rapidly quantify this cytotoxicity, a flow-cytometry test was developed based on the capacity of V tapetis to inhibit adhesion of clam hemocytes to plastic. Several bacteria:hemocyte ratios, the cytotoxicity of other Vibrio spp. pathogenic to bivalves, and that of various V. tapetis isolates were tested. Inhibition of adherence is detectable with as few as 5 bacteria per hemocyte. The greater cytotoxic activity of V tapetis compared to that of V splendidus and V pectenicida suggests a specific pathogenicity...

Regulatory T cells in dogs with multicentric lymphoma: peripheral blood quantification at diagnosis and after initial stage chemotherapy Arq. Bras. Med. Vet. Zootec.
Munhoz,T.D.; Anai,L.A.; Fonseca,D.M.; Semolin,L.M.; Sueiro,F.R.; Tinucci-Costa,M..
Lymphoma is the most common hematopoietic malignancy in dogs and one of the most frequent among all neoplastic diseases in this species. It can occur in several anatomical locations with distinct histological and immunophenotypes. Depending on the host immune response towards the tumor, prognosis information could be collected. Because its well established immunosuppressant, antitumor activity, the function of regulatory T cells (Tregs) in canine neoplasias has been investigated. In this study, we sought to quantify, using flow cytometry, the Tregs subpopulation in peripheral blood of healthy dogs (10) and in those diagnosed with type-B (14) and type-T (8) multicentric lymphoma before (at diagnosis) and after the first cycle (5-week) of 19-week...

Rhein induces apoptosis of human gastric cancer SGC-7901 cells via an intrinsic mitochondrial pathway BJMBR
Li,Yiwen; Xu,Yuqing; Lei,Bo; Wang,Wenxiu; Ge,Xin; Li,Jingrui.
Rhein is a primary anthraquinone found in the roots of a traditional Chinese herb, rhubarb, and has been shown to have some anticancer effects. The aim of the present study was to investigate the effect of rhein on the apoptosis of the human gastric cancer line SGC-7901 and to identify the mechanism involved. SGC-7901 cells were cultured and treated with rhein (0, 50, 100, 150, and 200 µM) for 24, 48, or 72 h. Relative cell viability assessed by the MTT assay after treatment was 100, 99, 85, 79, 63% for 24 h; 100, 98, 80, 51, 37% for 48 h, and 100, 97, 60, 36, 15% for 72 h, respectively. Cell apoptosis was detected with TUNEL staining and quantified with flow cytometry using annexin FITC-PI staining at 48 h after 100, 200 and 300 µm rhein. The percentage...

Seasonal to hour variation scales in abundance and production of total and particle-attached bacteria in the open NW Mediterranean Sea (0-1000 m) ArchiMer
Mevel, G.; Vernet, M.; Goutx, M.; Ghiglione, J. F..
We present the vertical and temporal dynamics of total vs. particle-attached bacterial abundance and activity over a 5 week period under summer to autumn transition in NW Mediterranean Sea. At a weekly time scale, total bacterial biomass and production in the euphotic layers was significantly correlated with phytoplanktonic biomass. At an hourly time scale, total bacterial biomass responded very rapidly to chlorophyll a fluctuations, suggesting a tight coupling between phytoplankton and bacteria for resource partitioning during the summer-autumn transition. In contrast, no influence of diel changes on bacterial parameters was detected. Episodic events such as coastal water intrusions had a significant positive effect on total bacterial abundance and...

Seasonal variation of stable isotope ratios of size-fractionated zooplankton in the Bay of Marseille (NW Mediterranean Sea) ArchiMer
Banaru, Daniela; Carlotti, Francois; Barani, Aude; Gregori, Gerald; Neffati, Nada; Harmelin-vivien, Mireille.
Stable isotope ratios of six size fractions of zooplankton (80 to >2000 mm) were analyzed seasonally in 2009-2010 at the SOMLIT site in the Bay of Marseille. Isotopic ratios generally increased with zooplankton size. The highest delta N-15 values were observed in the 1000-2000 mm fraction. The largest size class (>2000 mu m), dominated by gelatinous plankton, had lower delta N-15 values due to the low isotopic signatures of most of these organisms. In the larger size fractions (>1000 mu m), isotopic ratios were measured at the taxon level. Brachyuran, stomatopod, teleost and cephalopod larvae showed the highest delta N-15 values, and salps and pteropods the lowest ones. Lower values of both delta C-13 and delta N-15 were recorded in winter and...

Short-term interactions between phytoplankton and intertidal seagrass vegetation in a coastal lagoon (Bassin d'Arcachon, SW France) ArchiMer
De Wit, Rutger; Troussellier, Marc; Courties, Claude; Buffan-dubau, Evelyne; Lemaire, Emmanuelle.
We experimentally simulated the temporality of the interactions between intertidal seagrasses and phytoplankton in the context of nutrient enrichment from continental origin. Phytoplankton development was retarded after 1-day exposure to intertidal Zostera noltii Hornemann vegetation samples, with respect to control treatments. This was not explained by resource competition and we hypothesized a direct interference between seagrass leaves and phytoplankton. After separation from the vegetation in 5-day incubations, the final yield of large phytoplankton, mainly diatoms and haptophytes, was determined by nutrient loading. In contrast, Synechococcus-like cells (SYN), phototrophic picoeukaryotes and nanophytoplankton decreased after day 3, most likely due to...
